Components of naica® system

  • Shappire chips for 4 samples and 25 µl of PCR mix per reaction
  • Opal chips for 16 samples and 7 µl of PCR mix per reaction
  • Geode (sample partitioning and thermal cycling)
  • Prism (fluorescence reader)
  • Crystal Miner software

 

Easy to use digital PCR

Fast time-to-result (under 3 hours) with less than 15 mins of hands-on time

Crystals of 10.000 to 30.000 droplets are created from each sample using patented microfluidic technology

PCR amplification is performed immediately after crystal generation

Crystals are imaged to reveal the droplets that contain amplified targets in multiple channels

3-color detection capability (6-color available soon):

  • Prism3: Blue, Green and Red channel (for common fluorophores, e.g. FAM, HEX, Cy5 and more)

Software automatically counts the number of positive droplets in each population and converts it into absolute concentration of targeted DNA sequences.

The naica® system workflow

 

Reagents:

  • ready-to-use
  • two-component solution (buffer A and B)
  • available in 5x and 10x concentration
  • 10x concentration allows increased sensitivity for low concentration samples and rare targets 
  • excluding DNA primers, probes/intercalating dye and DNA template

 

1) naica® multiplex PCR MIX 

  • specially formulated for optimal TaqMan fluorescent probe-based Crystal Digital PCR™
  • high multiplexing capacity
  • contains an internal reference dye for partition detection and identification in the BLUE channel

 

2) naica® PCR MIX 

  • optimised for EvaGreen fluorescent intercalating dye-based Crystal Digital PCR™
  • Alexa Fluor 647 has to be added as reference dye to increase the basal fluorescence of droplets
